Thanks both! I’ve run the wheel/throttle with no load and there is no noise. I’ll buy a spoke wrench from Amazon and see how I land.

Broken spoke. Otherwise it's a internal issue with the hub.

Agreed, I would hoist the back end up and run the throttle on the hub to see if the noise persists. If it does it’s likely the hub, if not it’s probably the spokes. This is not financial advice.
